    Resveratrol attenuates chronic pulmonary embolism-related endothelial cell injury by modulating oxidative stress, inflammation, and autophagy

    Objectives: Due to Pulmonary Artery Endothelial Cell (PAEC) dysfunction, Pulmonary Hypertension (PH) persists even after the Pulmonary Embolism (PE) has been relieved. However, the mechanism behind this remains unclear. Method: Here, the authors incubated Human PAECs (HPAECs) with thrombin to simulate the process of arterial thrombosis. Results: CCK8 results showed a decrease in the viability of HPAECs after thrombin incubation. In addition, the expression of Tissue Factor (TF), Monocyte Chemoattractant Protein 1 (MCP-1), VCAM-1, ICAM-1, cleaved caspase 3, cleaved caspase 9, and Bax protein were all increased after thrombin incubation, while Bcl-2 was decreased. The effects of 3-MA treatment further suggested that autophagy might mediate the partial protective effects of Resveratrol on HPAECs. To observe the effects of Resveratrol in vivo, the authors established a Chronic Thromboembolic Pulmonary Hypertension (CTEPH) model by repeatedly injecting autologous blood clots into a rat's left jugular vein. The results exhibited that Mean Pulmonary Arterial Pressure (mPAP) and vessel Wall Area/Total Area (WA/TA) ratio were both decreased after Resveratrol treatment. Moreover, Resveratrol could reduce the concentration and activity of TF, vWF, P-selectin, and promote these Superoxide Dismutase (SOD) in plasma. Western blot analysis of inflammation, platelet activation, autophagy, and apoptosis-associated proteins in pulmonary artery tissue validated the results in PHAECs. Conclusions: These findings suggested that reduced autophagy, increased oxidative stress, increased platelet activation, and increased inflammation were involved in CTEPH-induced HPAEC dysfunction and the development of PH, while Resveratrol could improve PAEC dysfunction and PH

    AbstractTo realize automatic test of the shell motion parameters in the artillery experiment, the digital high-speed camera is used to gather the shell motion images. By applying digital image processing technique the characteristic images of shell are extracted accurately. Combined with the measuring principle of the shell motion parameters, a image test system is designed and realized automatic test of the shell motion parameters. The test results of shell motion images indicated that, the image extraction algorithm presented in this paper is easy to use and universal, and the precision of data measurement is increased. At present, the image test system has been applied in the data analysis in the artillery experiment and obtained good performance

    Reduced expression of Toll-like receptor 4 inhibits human breast cancer cells proliferation and inflammatory cytokines secretion

    <p>Abstract</p> <p>Background</p> <p>Tumor cell expression of Toll-like receptors (TLRs) can promote inflammation and cell survival in the tumor microenvironment. Toll-like receptor 4 (TLR4) signaling in tumor cells can mediate tumor cell immune escape and tumor progression, and it is regarded as one of the mechanisms for chronic inflammation in tumorigenesis and progression. The expression of TLR4 in human breast cancer cell line MDA-MB-231 and its biological function in the development and progression of breast cancer have not been investigated. We sought to characterize the expression of TLR1-TLR10 in the established human breast cancer cell line MDA-MB-231, and to investigate the biological roles of TLR4 in breast cancer cells growth, survival, and its potential as a target for breast cancer therapy.</p> <p>Methods</p> <p>TLRs mRNA and protein expressions were detected in human breast cancer cell line MDA-MB-231 by RT-PCR, real-time PCR and flow cytometry (FCM). RNA interference was used to knockdown the expression of TLR4 in MDA-MB-231. MDA-MB-231 transfected with the vector pGenesil-1 and the vector containing a scrambled siRNA were as controls. Recombinant plasmids named TLR4AsiRNA, TLR4BsiRNA and TLR4CsiRNA specific to TLR4 were transfected into human breast cancer cell line MDA-MB-231 with Lipfectamine™2000 reagent. TLR4 mRNA and protein expressions were investigated by RT-PCR, real-time PCR, FCM and immunofluorescence after silence. MTT analysis was performed to detect cell proliferation and FCM was used to detect the secretion of inflammatory cytokines in supernatant of transfected cells.</p> <p>Results</p> <p>The human breast cancer cell line MDA-MB-231 was found to express TLR1-TLR10 at both the mRNA and protein levels. TLR4 was found to be the highest expressed TLR in MDA-MB-231. TLR4AsiRNA, TLR4BsiRNA and TLR4CsiRNA were found to significantly inhibit TLR4 expression in MDA-MB-231 at both mRNA and protein levels as compared to vector control(vector transfected cells). TLR4AsiRNA mediated the strongest effect. Knockdown of TLR4 gene in MDA-MB-231 resulted in a dramatic reduction of breast cancer cell viability. The cytokines which were secreted by the TLR4 silenced cells, such as IL-6 and IL-8, also decreased significantly as compared with vector control. No significant difference was observed in siRNA control (Recombinant plasmid named ScrambledsiRNA transfected cells) compared to vector control.</p> <p>Conclusions</p> <p>These studies identified the expression levels of multiple TLRs in human breast cancer cell line MDA-MB-231 and demonstrated that knockdown of TLR4 could actively inhibit proliferation and survival of breast cancer cells. Taken together, our results suggest RNAi-directed targeting of TLR4 may be a beneficial strategy for breast cancer therapy.</p

    Seasonal dynamics of trace elements in sediment and seagrass tissues in the largest Zostera japonica habitat, the Yellow River Estuary, northern China

    Trace element accumulation is an anthropogenic threat to seagrass ecosystems, which in turn may affect the health of humans who depend on these ecosystems. Trace element accumulation in seagrass meadows may vary temporally due to, e.g., seasonal patterns in sediment discharge from upstream areas. In addition, when several trace elements are present in sufficiently high concentrations, the risk of seagrass loss due to the cumulative impact of these trace elements is increased. To assess the seasonal variation and cumulative risk of trace element contamination to seagrass meadows, trace element (As, Cd, Cr, Cu, Pb, Hg, Mn and Zn) levels in surface sediment and seagrass tissues were measured in the largest Chinese Zostera japonica habitat, located in the Yellow River Estuary, at three sites and three seasons (fall, spring and summer) in 2014–2015. In all three seasons, trace element accumulation in the sediment exceeded background levels for Cd and Hg. Cumulative risk to Z. japonica habitat in the Yellow River Estuary, from all trace elements together, was assessed as “moderate” in all three seasons examined. Bioaccumulation of trace elements by seagrass tissues was highly variable between seasons and between above-ground and below-ground biomass. The variation in trace element concentration of seagrass tissues was much higher than the variation in trace element concentration of the sediment. In addition, for trace elements which tended to accumulate more in above-ground biomass than below-ground biomass (Cd and Mn), the ratio of above-ground to below-ground trace element concentration peaked at times corresponding to high water discharge and high sediment loads in the Yellow River Estuary. Overall, our results suggest that trace element accumulation in the sediment may not vary between seasons, but bioaccumulation in seagrass tissues is highly variable and may respond directly to trace elements in the water column

    Cell-free measurements of brightness of fluorescently labeled antibodies

    Validation of imaging contrast agents, such as fluorescently labeled imaging antibodies, has been recognized as a critical challenge in clinical and preclinical studies. As the number of applications for imaging antibodies grows, these materials are increasingly being subjected to careful scrutiny. Antibody fluorescent brightness is one of the key parameters that is of critical importance. Direct measurements of the brightness with common spectroscopy methods are challenging, because the fluorescent properties of the imaging antibodies are highly sensitive to the methods of conjugation, degree of labeling, and contamination with free dyes. Traditional methods rely on cell-based assays that lack reproducibility and accuracy. In this manuscript, we present a novel and general approach for measuring the brightness using antibody-avid polystyrene beads and flow cytometry. As compared to a cell-based method, the described technique is rapid, quantitative, and highly reproducible. The proposed method requires less than ten microgram of sample and is applicable for optimizing synthetic conjugation procedures, testing commercial imaging antibodies, and performing high-throughput validation of conjugation procedures

    Preparation of Iron-nickel Alloy Nanostructures via Two Cationic Pyridinium Derivatives as Soft Templates

    In this paper, crystalline iron-nickel alloy nanostructures were successfully prepared from two cationic pyridinium derivatives as soft templates in solution. The crystal structure and micrograph of FeNi alloy nanostructures were characterized by X-ray diffraction, scanning electron microscopy and transmission electron microscopy, and the content was confirmed by energy-dispersive spectrometry. The results indicated that the as-prepared nanostructures showed slightly different diameter ranges with the change of cationic pyridinium derivatives on the surface. The experimental data indicated that the adsorption of cationic pyridinium compounds on the surface of particles reduces the surface charge, leading to an isotropic distribution of the residual surface charges. The magnetic behaviours of as-prepared FeNi alloy nanostructures exhibited disparate behaviours, which could be attributed to their grain sizes and distinctive structures. The present work may give some insight into the synthesis and character of new alloy nanomaterials with special nanostructures using new soft templates
